Introduction

It is driven by a rotary motor, which usually consists of a motor, a reducer, a power transmission shaft and a rotary clamping device. Among them, the motor provides power, the reducer converts the high-speed rotation of the motor into appropriate low-speed rotation, the power transmission shaft transmits the rotation speed to the rotating clamping device, and the rotating clamping device transmits this rotation force to the rotating flask.

rotovap3

The rotary clamping device usually consists of two clamping arms and clamping claws. The clamping claws clamp the flask in the middle position, and the clamping arms are connected with the power transmission shaft, so that the power of the rotary motor can be transmitted to the rotary flask through the clamping arms, thus realizing the rotation of the flask.

During the experiment, the rotating speed of the rotary flask needs to be adjusted according to the experimental requirements, generally between 100 and 300 rpm. The increase of rotating speed can increase the surface area of the solution and promote the evaporation of the solvent, but too fast rotating speed can also cause the sample to splash or the beaker to tilt, so it needs to be adjusted according to the actual situation.

The hot oil used for heating the rotary evaporator equipment is high-temperature thermal conductivity oil. This thermal conductivity oil has high thermal stability and thermal conductivity, is suitable for high-temperature heating system, and can stably transfer heat at higher temperature.

Common high-temperature thermal conductivity oils include: diphenyloxy diphenyloxy phosphine (Dowtherm A), triphenyloxy triphenylphosphine (Dowtherm J), Diphenyl ether, etc.

operating principle

● Heating system:
Rotovap is usually equipped with a water bath or oil bath as a heating system, which heats the solvent in the sample bottle to evaporate. The heating temperature can be precisely adjusted by microcomputer PID control system to meet the evaporation needs of different solvents.
● Rotating system:
The bottle is mounted on a rotating shaft and rotated by a motor drive. The rotation speed can usually be adjusted within a certain range (such as 10-140rpm or 20-200rpm, etc.), depending on the device model and experimental requirements. The rotating motion helps to increase the evaporation area of the solvent on the inner wall of the sample bottle, thus improving the evaporation efficiency.
● Condensing system:
The evaporated solvent vapor is condensed by a condenser. The condenser usually adopts a serpentine coil structure (such as straight or inclined) to increase the condensing area and condensing efficiency. The cooling medium in the condenser (such as water or refrigerant) absorbs the heat of the steam by circulating it and condenses it into a liquid.
● Vacuum system:
The Rotovap is usually connected to a vacuum pump to create a negative pressure environment inside the device. The negative pressure environment helps to reduce the boiling point of the solvent, thus achieving rapid evaporation of the solvent at lower temperatures. In addition, the vacuum environment can also reduce the oxidation and decomposition of solvents during evaporation.
● Collection system:
The condensed solvent liquid flows into the recovery bottle for collection. Recovery bottles usually have sufficient capacity to contain the solvent that evaporates throughout the experiment.

Operation Knowledge

Control the sample amount

  • According to the needs of the experiment, accurately measure the required amount of samples, and use appropriate tools (such as burette and syringe) to add samples.
  • Control the sampling speed to avoid too fast or too slow sampling speed. The sampling speed can be controlled by manually adjusting the sampling speed or using an automatic sampling device.
  • Leave a certain space before adding samples to avoid solution overflow. According to the nature of the sample and the volume of the container, reasonably determine the sample addition amount to avoid exceeding the bearing capacity of the container.

Observe the state of the solution

  • During the experiment, observe the state of the solution at any time, especially during the sample adding process. If it is found that the solution is close to the overflow edge, stop the sample addition immediately and adjust the rotation speed or reduce the sample addition.
  • Pay attention to the tightness of the flask, ensure the tight connection between the flask and the rotating clamping device, and avoid solution leakage.

Adjust according to experimental experience

  • According to the previous experimental experience and results, gradually master the appropriate rotation speed and sample addition to avoid solution overflow or waste.
  • If overflow occurs during the experiment, it should be recorded and adjusted in time to improve the accuracy of the next experiment.

Test Method

Chromatography

  • Gas Chromatography (GC): It is suitable for detecting the evaporation effect of volatile substances, such as volatile organic compounds. Samples before and after evaporation were collected and analyzed by gas chromatograph. • Liquid Chromatography (LC): It is suitable for detecting the evaporation effect of non-volatile substances, such as water-soluble compounds. Samples before and after evaporation were collected and analyzed by liquid chromatography. Compare the changes of peak area or peak height of substances before and after evaporation to evaluate the evaporation effect.

Spectroscopy

  • Ultraviolet-visible spectrum (UV-Vis): It is suitable for detecting the evaporation effect of absorbent substances in solution when using a 5 liter rotovap. Samples before and after evaporation were collected, and the absorption spectrum was measured by UV-Vis spectrometer. Compare the changes of absorbance before and after evaporation to evaluate the evaporation effect.
  • Infrared spectrum (IR): It is suitable for detecting the evaporation effect of functional groups in solution. Samples before and after evaporation were collected, and the infrared spectrum was measured by infrared spectrometer. Compare the changes of peak intensity or position before and after evaporation to evaluate the evaporation effect.
